CF has long been associated with lung disease but can also severely affect gastrointestinal tissues. CF carriers̶those with a CF-predisposing genetic mutation̶have several CF-related symptoms, including constipation. In addition, some of the newer drugs for chronic constipation have a mechanism of action that stimulates CFTR function, suggesting that CFTR function is closely related to the development of constipation. However, there have been no recent studies on whether CFTR dysfunction causes constipation. In this study, to examine whether inhibiting CFTR function in the intestine induces constipation, we fed mice food containing CFTRinh172, a CFTR-specific inhibitor. The results showed that the fecal water content (22.49%) was 74% of that of the control group (30.45%; p = 0.15 vs. the control group) on day 13 after inhibitor administration. After switching to the normal diet, the average fecal water content recovered to almost the same level as that of the control group on day 3. There was a significant decrease in fecal water content on day 13 of the inhibitor treatment compared to day 5 after diet switching, when fecal water content recovery was stable (30.50%; p < 0.05). Furthermore, the fecal transit time was prolonged from 26 to 30 hours in the inhibitortreated mice compared to 24 hours in the control mice. In conclusion, constipation was induced in mice that received approximately 20 μg of CFTRinh172 daily for 13 days.
